Title:The influence of resistant soybean (PI 227687) foliage and coumestrol on the metabolism of xenobiotics by the soybean looper, Pseudoplusia includens (Walker).

Abstract:The metabolic activity of the noctuid Pseudoplusia includens [Chrysodeixis includens] towards several xenobiotics was determined by rearing larvae on susceptible (Bragg) and resistant (PI 227687) soyabean cultivars. Midgut homogenates from last-instar larvae reared on PI 227687 throughout their larval stages had a higher level of glutathione transferase and monooxygenase activity (1.5- and 2-fold, resp.) than those reared on Bragg leaves. Minor metabolic differences between larvae reared on the 2 cultivars were observed for NADPH cytochrome c reductase [NADPH-cytochrome-c2 reductase] activity and the hydrolysis of p-nitrophenyl acetate, α-napthyl acetate and acephate. There was no increase in the level of glutathione transferase activity of last-instar larvae fed PI 227687 foliage for a short period (48 h). The level of monooxygenase activity was also less than previously observed. Small decreases in p-nitrophenyl acetate and α-napthyl acetate activities were also associated with short-term feeding on PI 227687 foliage. The incorporation of coumoestrol, an allelochemical associated with insect resistance in PI 227687 foliage, into the larval artificial diet significantly increased the level of glutathione transferase activity. None of the other enzymes assayed were affected by the coumoestrol-amended diet. The increase in glutathione transferase activity in larvae fed the coumoestrol-amended diet relative to the untreated control diet was similar to that observed when the larvae were fed PI 227687 foliage versus Bragg foliage. It is concluded that coumoestrol, and other factors, contribute towards the resistance of PI 227687
