Title:First report of transmission of Soybean mosaic virus and Alfalfa mosaic virus by Aphis glycines in the New World

Abstract:To determine if endemic isolates of soybean mosaic virus (SMV) and alfalfa mosaic virus (AMV) could be transmitted by the introduction of soyabean aphid (Aphis glycines) in the north central region of USA, transmission experiments were conducted using 3, 5 and 10 aphids per soyabean (cv. Williams 82) plant. For SMV experiments, the aphid-transmissible field isolate A15 (GenBank Accession no. AF242844) and as a negative control, the non-aphid transmissible isolate N (GenBank Accession no. D500507), were used. For AMV experiments, a field isolate of AMV was used. Myzus persicae and A. glycines were maintained on broad bean [faba bean] and virus-free soyabean, respectively. No transmission of SMV-N occurred in any of the tests. For the A15 isolate, 30, 50 and 50% transmission efficiencies (TEs) were obtained with 3, 5 and 10 M. persicae, respectively, per plant; while 30, 40 and 40% TEs were obtained with 3, 5 and 10 A. glycines, respectively, per plant. Transmission levels of both aphids did not differ significantly. For AMV, corresponding TEs were 0, 0 and 20%. The results suggest that the introduced A. glycines can be an efficient SMV vector, but a less efficient AMV vector.
